Structure of an Escherichia coli N-acetyl-d-neuraminic acid lyase mutant, E192N, in complex with pyruvate at 1.45 Å resolution
The structure of a mutant variant of Escherichia coli N-acetyl-d-neuraminic acid lyase (NAL), E192N, in complex with pyruvate has been determined in a new crystal form. It crystallized in space group P212121, with unit-cell parameters a = 78.3, b = 108.5, c = 148.3 Å. Pyruvate has been trapped in the active site as a Schiff base with the catalytic lysine (Lys165) without the need for reduction. Unlike the previously published crystallization conditions for the wild-type enzyme, in which a mother-liquor-derived sulfate ion is strongly bound in the catalytic pocket, the low-salt conditions described here will facilitate the determination of further E. coli NAL structures in complex with other active-site ligands. Dropping your habit of drinking sugary, caffeinated beverages like Coke and Pepsi could make you feel better, research suggests. Cross-national epidemiologic studies have identified a possible link between sugar intake and rates of major depressive disorders. In addition, when researchers took a look at the diets of people with depression, they found that their sugar consumption was much higher than the general population. It wasn't just sugar that seemed to be a culprit, however. One small cohort trial found that when people eliminated sugar and caffeine from their diet those with unexplained depression experienced improved moods within a week. When caffeine and sugar were reintroduced into their diet, their symptoms returned; but, placebo did not produce the same effect. Giving further credence to the caffeine-depression link, another study found that regular consumption of high levels of caffeine (>750 mg daily) appears to be associated with depression. Teens With Depression, Other Mental Disorders May Be at Risk for Internet Addiction
According to Taiwanese researchers, teens with various mental disorders, such as depression and ADHD (attention deficit hyperactivity disorder), may be more at risk for becoming addicted to using the Internet. The researchers asked 2,293 seventh graders to fill out questionnaires regarding whether they had ADHD, depression, social phobia or abnormal feelings of hostility. Six to 24 months later, they were asked about their Internet usage, such as how many hours they spent on the Internet and what types of sites they visited. The researchers found that boys and girls with ADHD or hostility were more likely to be addicted to the Internet than those without those conditions. Social phobia and depression was associated with Internet addiction in girls, but not boys. At this time, there is no universally agreed upon definition for Internet addiction, but it is generally considered to be excessive use which negatively impacts grades, relationships or the person's emotional state.
